Emerging drugs for heart failure
- Oddelenia zlyhávania a transplantácie srdca, NÚSCH, a.s., Bratislava, Slovenská republika
Pharmacological treatment of heart failure has changed only very slightly during the last ten years. From the large number of drugs that were studied for the treatment of chronic heart failure only ivabradin has attained clinical use. Currently, there are tens of new molecules in various stages of development. Most is expected from the research on new drugs for the treatment of acute heart failure and heart failure with preserved left ventricular ejection fraction. For these patients or clinical situations we still do not have drugs that have scientifically been shown to be effective, particularly in terms of positive impact on patients' survival. In the near future approval for clinical usage of LCZ696 is pending. LCZ 696 is a combination of the angiotensin receptor blocker valsartan and neprilysin inhibitor. The extensive clinical trial (PARADIGM-HF) has shown that LCZ696 in patients with chronic heart failure based on left ventricular systolic dysfunction significantly reduces the number of deaths from cardiovascular causes, or hospitalization for heart failure. The study was stopped prematurely at a median follow-up of 2.7 years because of the clear benefit from the use of LCZ696. In this review article substantial information on other substances such are serelaxin, guanylcyklase stimulators, natriuretic peptide analogues, omecamtiv mecarbil, albiglutide, poloxamer 188 and istaroxime is also summarized Fig. 3, Tab. 1, Ref. 18, Online full text (Free, PDF)
