The effect of PPAR? agonist - pioglitazone on hypertension and redox regulation in young and adult SHR

Aim: The PPARγ is an uclear receptor that takes part in the regulation of energy homeostasis, antioxidant response and hypertension development and can be also activated using an exogenous agonist such as pioglitazone. The aim of this study was to determine the influence of pioglitazone (PIO) treatment on blood pressure (BP) and redox systems in the brainstem (BS) and the left heart ventricle (LV) and specify changes among young and adult spontaneously hypertensive rats (SHR).

Methods: Blood pressure was determined by the plethysmography method. Production of superoxide radicals was detected by Lucigenin-enhanced chemiluminiscence. Quantitative RT-PCR was used for gene expression and detection. Lipid profile and superoxide dismutase (SOD) activity was determined spectrophotometrically, using specific assays.

Results: Administration of PIO to SHR slowed blood pressure increase and improved lipid profile in young, but not in adult' rats. In BS of young rats we observed increase of PPARγ and SOD2 gene expression without SOD activity changes, while in LV we found SOD activity increase without changes in PPARγ expression and without influence on superoxide production.

Conclusion: It can be concluded that PPARγ agonist - PIO directly affected redox regulation in LV, while in BS redox regulation correlated with PPARγ activation. Influence of PIO treatment was more effective in regulation of hypertension and redox response in young SHR, compared with adult, animals. Experimental data suggests different influence of glitazones according to the age, the type of agonist and/or the experimental model of hypertension.
