The effect of neuronal NO-synthase inhibition on the cardiovascular system of Wistar rats from the prenatal period to adulthood

Aim: The long-term decrease of nitric oxide (NO) production evokes serious alterations in the cardiovascular system. The aim of this study was to investigate the long-term effect of neuronal NO-synthase (nNOS) inhibition with 7-nitroindazole (7-NI) on blood pressure, heart, kidneys, thoracic aorta and carotid artery of normotensive Wistar rats from the prenatal period to early adulthood.
Methods: In the experimental group 4-week-old males (n=10) were treated with 7-NI (10 mg/kg/day, in drinking water) for six weeks. The animals were offspring of mothers who had been administered 7-NI (10 mg/kg/ day) in drinking water from two weeks before pregnancy until end of weaning. Age-matched animals (n=10) were taken into control group. Blood pressure was measured weekly noninvasively by the plethysmographic method on the tail artery. After perfusion with a glutaraldehyde fixative (120 mmHg), the heart and kidneys were weighed and the thoracic aorta and carotid arteries were processed for morphological investigation.
Results: Long-term administration of 7-NI resulted, in spite of unchanged blood pressure and body weight compared with the control group, in decrease of heart and kidney weight. Morphological investigation of the vessel wall (tunica intima+media) of experimental rats revealed a decrease of the wall thickness, cross sectional area and wall thickness/inner diameter ratio, without affecting the inner diameter in the both conduit arteries. Circumferential tension of thoracic aorta and carotid artery was increased.
Conclusion: Chronic NO deficiency due to the inhibition of nNOS from prenatal period to adulthood resulted in pressure-independent hypotrophy of the heart, kidney and arterial wall of conduit arteries in normotensive Wistar rats.
