Characteristics of erythrocytes and risk factors for cardiovascular diseases

Deformability refers to an ability of red blood cells (RBCs) to change their shape in order to pass through capillaries in circulation. Deterioration in RBC deformability contributes to alterations in microcirculatory blood flow and delivery of oxygen to tissues. Decreased deformability of RBC has been found to be the marker of adverse outcomes in cardiovascular diseases (CVD). One of factors responsible for maintenance of RBC deformability is the Na,K-ATPase known as crucial enzyme in maintenance of intracellular ionic homeostasis. Changes in concentration of ions affect cellular volume and consequently RBC deformability. This review summarizes the knowledge concerning the RBC deformability in connection with selected risk factors of CVD, including hypertension, hypercholesterolemia, and diabetes mellitus, based on papers from human studies. We attempted to provide an update on important issues regarding the role of Na,K-ATPase in RBC deformability. The parameters of RBCs are modified not only in CVD, but already in risk factors of CVD before clinical manifestation of disease. Data presented in this review allow us to suggest that in patients suffering from hypertension as well as diabetes mellitus the Na,K-ATPase appears to be responsible for the changes leading to alterations in RBC deformability. The triggering factor for changes of RBC deformability during hypercholesterolemia seems to be the increased content of cholesterol in erythrocyte membranes that modified its mechanical properties.
